Abstract本发明涉及领域,具体涉及一种动物细胞用表达载体,由上游起依次含有强表达诱导性启动子、基因整合用多克隆位点、核糖体进入位点序列、二氢叶酸还原酶基因、不含增强子的SV40早期启动子基因、neo基因和抗药性基因。本发明还提供了上述表达载体的制备方法、重组载体及应用。本发明的表达载体中用不含增强子的启动子弱化neo基因的表达,dhfr基因作为共扩增基因位于核糖体进入位点IRES的下游,降低dhfr基因的翻译效率,结合G418加压筛选和二氢叶酸还原酶基因压力筛选,增加目的蛋白的表达量。
