Abstract	A pair of volatile isomers, cyperotundone and α-cyperone, were successfully preparatively separated and purified from the rhizomes of Cyperus rotundus Linn. by high speed counter-current chromatography (HSCCC) with two-phase solvent system consisting of n-hexane-acetonitrile-acetone-water (7:6:0.5:1.5, v/v). 105.5 mg of cyperotundone with purity over 95% and 117.4 mg of α-cyperone with purity over 99% were obtained from 2.0 g of the essential oil of C. rotundus, respectively.
