Objective: To compare the inhibitory activities against ovarian cancer HEY cells of the extracts and fractions of Panax ginseng, Panax quinquefolium, Panax japonicas and Panax notoginseng. Methods: Water and ethanol extracts were prepared by reflux extraction with water and ethanol. Total saponins and their 20(S)-protopanaxatriol saponins (PTS) and 20(S)-protopanaxadiol saponins (PDS) fractions were prepared using macroporous resin chromatography. MTT assay was used to test the anti-proliferative activity of these extracts against HEY ovarian cancer cells. PI staining was used to detect the cell cycle distribution and western blot was performed to detect the expression of cell cycle related proteins such as CDK4, CDK6, and cyclin D1. In addition, wound healing assay was conducted to detect the effect on migration of indicated extracts. Results: All the indicated extracts showed varied activities against HEY cells. Among them, PDSPQ, TSPJ, EEPG, TSPG, and PDSPG showed relative higher inhibitory rate with 45.59%, 45.78%, 50.48%, 46.98%, and 64.36%, respectively. Further, PDSPQ, TSPG, and PDSPG could down-regulate the expression of CDK4, CDK6, and cyclin D1, while had no effect on the migration of HEY cells. Conclusion：The fractions with PDS showed more effective on inhibiting HEY .
