Abstract	Dimethyl sulfoxide (DMSO) is a prevalent solvent for dissolving drugs and cryo-preservation of cells. In our previous study, we observed that cancer cell lines and human primary tumor dissociated cells responded diversely to DMSO on chip. Although cytotoxicity of DMSO have been known for decades, it has always been used as normalization controls during in vitro drug screens. Thus, DMSO cytotoxicity might have been overlooked. Furthermore, no synergy between DMSO and drugs could be documented with conventional drug screening assays. Hence, in this study, we have tested the effect of DMSO against multiple cancer cell lines from diverse tissue origins. By utilizing the microfluidic chip assay that we developed, the cytotoxic effect of DMSO that was long known but ignored during in vitro drug screening will be discussed. On the other hand, we have also tested the effect of DMSO on the mixture of 2 cancer cell lines. Results paved the way for further investigation on the effect of anti-cancer drugs. By comparing with corresponding results of pure cell populations, we plan to investigate the potential cell-cell interactions that might affect drug specificity and efficacy in mixed cell populations. Results will shed light on the impact of intra-tumor heterogeneity on drug response and provide insight to designing personalized cancer therapy.
