Abstract	Noncompetitive antagonists of N-methyl-D-aspartate (NMDA) type glutamate receptors, such as MK-801 are neuroprotective, but paradoxically produce excitation and toxicity in neurons in the limbic cortex which includes the entorhinal cortex. We have investigated the effects of systemic administration of MK-801 (5 mg/kg; 4 hours) on genes expressed in the entorhinal cortex utilizing differential screening of cDNA libraries, pBluescript libraries were constructed from mRNAs isolated from saline (control) and MK-801 treated rat entorhinal cortices. More than 1000 cDNAs were randomly picked from the treated library, and duplicate southern blot filters prepared, and differentially probed with p labelled cDNAs generated from the reverse transcription of mRNAs from control and MK-801 treated rat entorhinal cortices. This reverse northern screening procedure revealed 16 candidate cDNAs which are regulated by MK-801. Efforts to confirm the up or down regulation, and regional specific expression of these cDNAs by in situ hybridization are currently underway. The sequence and identification of such genes should provide insights to the mechanism(s) by which noncompetitive NMDA receptor antagonists produce paradoxical toxicity.
