Glucomannan Biomaterials Targeting C-type Lectin Receptors For Drug Delivery And Tissue Regeneration | |

Abstract | C-type lectin (CLEC) is a family of immune cell receptors that can bind carbohydrate molecules and mediate both innate and adaptive immune responses. Interactions between carbohydrates and major CLECs initiate endocytosis and other cellular events that are critical for tissue regeneration. In our study, we have developed two glucomannan polymers into three biomaterial types (oxBSP, acBSP and sKGM) in the forms of nanoparticles and injectable gels. We evaluated the performance of oxBSP as drugdelivery vehicles in both in vitro and in vivo models, assessed the potential of acBSP as tissue engineering scaffolds, and tested the ability of sKGM in promoting wound healing in situ. Our results suggested that the oxBSP-based nanoconjugates exhibited high affinity for macrophage mannose receptors, acBSP supported embryonic stem cells adhesion, and sKGM had the potential to sequester nutrient proteins in the wound fluid and consequently enhance wound healing. Our series of studies reveal the high potential of exploiting the interactions between CLECs and glucomannan-based biomaterials for drug delivery, tissue engineering and regenerative medicine. |
