Abstract本发明公开了一种用于筛选抗血管损伤药物的斑马鱼血管损伤模型及其建立方法和应用。本发明通过血管生长因子受体抑制剂对受精后的斑马鱼胚胎进行处理,得到血管损伤的斑马鱼,建立一个斑马鱼血管损伤模型,而后再用检测的药物处理斑马鱼1-3天,通过形态学观察以及实时定量PCR检测斑马鱼组织中血管内皮生长因子受体(Flk-1A,Flk1-B,Flt1)基因表达的水平来判断检测的药物对斑马鱼血管损伤的治疗效果。试验结果表明,本发明所建立的模式生物斑马鱼血管损伤模型,其造模率极高,可以模拟体内血管新生不足的环境进行快速筛选具有促血管新生类的药物,效果易于观察明显,在实验过程中用药量低,实验结果准确可重复性强。该模型可以成批量筛选潜在的药物,工作效率高。
