Abstract | 本发明公开了GSK3β抑制剂和/或HRR抑制剂在制备治疗FHIT缺失型肺癌的药物中的应用。本发明首次发现糖原合成酶激酶3β(GSK3β)和同源重组DNA 修复(HRR)通路是肺癌中FHIT的合成致死标靶;抑制GSK3β或HRR通路,包括ATR/BRCA1/RAD51轴,可显著抑制FHIT缺失肺癌细胞中的DNA双链断裂修复,导致DNA损伤累积和细胞凋亡;为GSK3β和HRR通路作为FHIT缺失型肺癌可行药物靶点提供了依据。而且,本申请进一步研究了在非FHIT缺失肺癌中,同时抑制FHIT和GSK3β/ HRR,仍会导致细胞产生合成致死,进一步扩展了应用方向。 |
