Abstract | Filamentous fungi have the remarkable ability to undergo asexual reproduction, generating an enormous quantity of spores (a.k.a. conidia). These spores serve as the primary means of dissemination, acting as infectious propagules for human and plant pathogens, as well as significant contributors to food spoilage. The germination of spores and their subsequent growth as hyphae under favorable conditions are critical processes for infection and food spoilage. Therefore, understanding the mechanisms underlying conidiation and spore germination holds not only biological importance but also clinical and agricultural implications. To gain insight into the molecular mechanisms of conidiation and spore germination, we performed active transcription profiling by ChIP-seq and transcriptome profiling by RNA-seq on Aspergillus nidulans at distinct stages of conidiation and germination with a high temporal resolution. Our results unveiled dynamic transcriptional and post-transcriptional changes throughout different stages of germination and conidiation. Each stage exhibited enrichment of distinct gene sets with specialized functions. For some of these gene sets, we have identified potential candidate transcriptional regulators that could serve as valuable drug targets for impeding conidiation and spore germination. Further characterization of these regulators holds great promise for the development of novel antifungal drugs for preventing human and plant fungal infections. |
