天然香豆雌酚類化合物的藥理活性和作用機制

Abstract天然香豆雌酚是一類源自植物(主要是豆科植物)的多環芳香族次級代謝產物,其化學結構由香豆素部分和苯並呋喃部分共用一個C=C鍵的含氧四環雜環化合物。近年來,香豆雌酚類化合物,特別是擬雌內酯、蛇床子素、補骨脂素、甘草次酸和甘草酚對多種疾病的藥理活性和藥用潛力日益受到關注。已發現有120餘種香豆雌酚類化合物,大致可分為含核心骨架香豆雌酚、二甲基吡喃香豆雌酚、呋喃香豆雌酚、O-糖基化香豆雌酚及其他類型。香豆雌酚類化合物具有廣泛的藥理活性,包括雌激素活性、抗癌、抗炎、抗骨質疏鬆、器官保護、神經保護、抗糖尿病、抗肥胖、抗菌、免疫抑制、抗氧化和皮膚保護等。我們從煙豆(Glycine tabacina (Labill.) Benth)中分離得到8個新香豆雌酚化合物glytabastan A-H,其中glytabastan B通過抑制MAPK和PI3K/AKT信號通路而顯著改善小鼠2型膠原蛋白誘導的類風濕關節炎症狀。此外,我們還發現甘草酚(glycyrol)和菜豆醇(phaseol)通過抑制破骨細胞分化成熟、促進成骨細胞形成的雙向調節功能而顯著改善脂多糖(LPS)誘導的小鼠骨質疏鬆症狀,提示這些香豆雌酚類化合物在預防或治療類風濕關節炎和骨質疏鬆症的潛在應用價值。
