A novel interaction between CtBP and TRIM28: implication for the development of breast cancer

AbstractC-terminal Binding Proteins (CtBP) and Tripartite motif-containing 28 (TRIM28) 28 are both important transcription co-regulators and have been proved that involved in many important biological processes in both tumor and normal cells. The co-expression of TRIM28 and CtBP has been found in many organs, such as lung, liver, ovarian and breast. Based on the previous studies, the expression levels of TRIM28 and CtBP are closely related to tumor aggressiveness, poor prognosis, and worse survival. However, the function of the complex formed by interaction between TTRIM28 and CtBP in the progression of breast cancer or any other types of cancer cells has still not been elucidated. In our study, TRIM28 and CtBP play similar roles ofin promoting breast cancer cell migration and tumorigenesis in vitro. Mechanically, TRIM28 directly interacts with CtBP1&2, and co-localizes in nucleus. The decreased level of CtBP showswas observed in TRIM28 knock down and knock out cell. In addition, we proved that TRIM28 protects CtBP1&2 from ubiquitination mediated degradation but not through the transcription level regulation. More interesting is that CtBP1&2 knock down can also reduce the protein level of TRIM28 by inducing autophagy, the results above indicate that TRIM28 and CtBP1&2 can form the complex and protect each other from degradation viathrough different pathways. After retrieving data from public databases, breast cancer patients with high levels of TRIM28 and CtBP1&2 indicate poor outcomes, whereas patients with low TRIM28 and CtBP1&2 expression have the improved prognosis. Taken together, this study providess a novel interaction between TRIM28 and CtBP, and the complex of TRIM28-CtBP may establish serve as a promising target for the therapy of breast cancer in the future.
