Abstract | Depression is a complex mental illness that severely threatens human health. At present, clinical antidepressant drugs generally have some obvious drawbacks such as slow onset, low efficacy, and serious adverse reactions. Traditional Chinese medicine is characterized by multiple components, multiple targets, and minimal toxic side effects, which has marked strengths in the treatment of depression. Especially, in clinical observations, the combination of ZY01 Granules and SSRI antidepressants was found to be able to significantly accelerate the onset time, improve the effective rate, and reduce adverse reactions, thus indicating that ZY01 Granules may effectively improve depressive symptoms and exert antidepressant pharmacological activity. In this study, corticosterone-induced PC12 cell injury model was used to simulate the pathological process of glucocorticoid induced neurological dysfunction. Cell survival rate was measured with the CCK8 method, and apoptosis was detected by Hoechst 33342 staining method, using Annexin V- FITC/PI flow cytometry to evaluate the neuroprotective effect of ZY01 Granules. The results showed that ZY01 Granules (12.5, 25, 50 μg/ml) can effectively improve the survival rate of damaged PC12 cells induced by corticosterone, and exert antidepressant activity via inhibiting cell apoptosis. |
