Abstract | Objective Nitric oxide(NO) is implicated in brain injury and early neurological deterioration in acute ischemic stroke. In present study, we investigated the protective effects of Artemisinin against sodium nitroprusside(SNP)-induced oxidative injury in PC12 cells and cortical neurons as well as exploring its underlying mechanism. Methods Cells were treated with SNP to create a neuronal injury model. The protective effect of Artemisinin was evaluated by pre-treatment of PC12 cells with Artemisinin before exposure to SNP. The cell viability and nuclear morphology were evaluated using MTT assay or Hoechst staining respectively. Activation of protein kinase B (Akt) and extracellular regulated protein kinase (ERK) was determined by Western blotting analysis. The kinase pathway involved in the effect of Artemisinin was studied by kinase inhibitors. Results Artemisinin significantly attenuated SNP-induced oxidative injury in PC12 cells and primary cortical cultured neurons by stimulating the activation of ERK but that of AKT. Moreover, inhibition of ERK with the ERK pathway inhibitor, PD98059 blocked the protective effect of Artemisinin while the PI3K inhibitor, LY294002 had no effect.
