AbstractCryptotanshinone (CTS) is one of the active ingredients of the Salvia miltiorrhiza. It was found most recently that CTS significantly inhibits the progression of leukemia, glia tumor and gastric carcinoma. These findings indicated the anti-tumor effect of CST which has attracted more and more attention. However, the anti-tumor mechanism of CST remain to be elucidated. In this study, we found that CST attenuates the tyrosine phosphorylation of IGF-IR, a tyrosine kinase receptor overexpressed in various cancer tissues, and its downstream signaling induced by IGF-1in PC12 cells, a pheochromocytoma of the rat’s adrenal medulla and glia tumor cell line. The suppression of the expression of phosphatase SHP-2 by siRNA reversed the inhibitory effect of CST on the phosphorylation of IGF-1R while knockdown of other phosphatases involved in IGF-1R tyrosine phosphorylation had no effect. These results indicated that the inhibitory effect of CST was mediated by SHP-2. Consisitent with this hypothesis, CST time-and concentration-dependently increased the phosphorylation of SHP-2 and the enzyme activity of SHP-2. These results putting together, suggest that CST-1 via the stimulation of phosphatase SHP-2 in neuronal cells. Acknowledgement: Partly supported by the organizers of the conference.
