AbstractAge-related macular degeneration(AMD) is a leading cause of severe visual loss and legal blindness in the aging population. Recent studies have indicated that oxidative damage to the retina plays a key role in the pathogenesis of AMD. In this study, Artemisnin, a famous anti-malarial drug isolated from the Chinese plant, Artemisia annua, was found to suppress dramatically hydrogen peroxide-induced oxidative damage in retinal neuronal cells, RGC-5 cells. It is found that Artemisnin concentration-dependence attenuates the accumulation of intracellular reactive oxygen species(ROS) and the increase of the mitochondrial membrane potential induced by hydrogen peroxide. Western blot analysis showed that hydrogen peroxide leads to the reduction of phosphorylated p38 and ERK1/2, whereas Artemisinin stimulates the phosphorylation of p38 and ERK1/2 and reverses the inhibitory effect of hydrogen peroxide on these two kinases. Consistent with the effect of Artemisinin is inhibited by the p38 kinase inhibitor, PD16 and ERK1/2 kinase pathway inhibitor, PD98059. Taken together, these results indicated that Artemisinin protects hydrogen peroxide-induced RGC-5 cells death by modulating the p38 and ERK1/2 pathways.
