Abstract

神經分化是神經發育過程中的一個重要環節,正常的神經分化對於神經系統功能的建立具有重要意義。神經分化的過程受到許多外在和內在因素的調控。生長相關蛋白43(Growth Associated Protein 43 (GAP-43), 也叫F1, B-50, pp46和neuromodulin)是一種神經特異性蛋白質,位於神經軸突膜上,參與神經細胞外生長及突觸發育形成和神經細胞再生,是神經細胞內一個重要的調控神經分化的內在調控因子 FoxO3a 是FoxO转录因子家族的成员之一,广泛参与细胞的增殖、分化、代谢及凋亡的调控。最近研究证明FoxO参与了多种非神经细胞分化的调控,但是FoxO转录因子对细胞分化的影响依参与的FoxO家族成员不同而作用不同,例如FoxO1抑制脂细胞和成肌细胞的分化,而FoxO3a则促进血细胞的分化,就同一种家族成员而言,作用的组织不同, 细胞不同,其对细胞分化的影响亦不一致。例如,FoxO3a可以促进红细胞的分化,但在心肌细胞中,FoxO3a则可以抑制心肌细胞的分化。这些研究显示FoxO对细胞分化的影响具有家族成员特异性、组织特异性及细胞特异性。FoxO3a对非神经细胞分化的影响已有报道,但FoxO3a对神经细胞分化的作用及作用机制则不是很清 本项目研究了FoxO转录因子对神经细胞分化, 轴突生长相关蛋白GAP-43的作用及机制。
