AbstractDepression is a prevalent psychiatric disorder and a leading cause of disability worldwide. Despite variety of available treatments currently being used in the clinic, but a substantial proportion of patients is unresponsive to them, urging the need to develop more effective therapeutic approaches. Hederagenin, is a triterpenoid saponin firstly found in ivy (Hedera helix L.), demonstrates several biological activities, including anti-apoptosis, anti-hyperlipidemic, anti-inflammatory properties, and has been shown to have potential therapeutic roles in the depression, however, the role of hederagenin in depression has not been elucidated. Chronic and high dosages of corticosterone treatment are able to stimulate anxiety- and depression-like behavioral changes in the behavior, neurochemistry and brain morphology such as the reduction of hippocampal cells. In this present study we reported that corticosterone-induced damage to PC12 cells, which can be ameliorated by hederagenin treatment. Specifically, hederagenin prevented the decline of mitochondrial membrane potential, reduced intracellular ROS production and decrease apoptosis induced by corticosterone. Moreover, hederagenin neuroprotective effect is likely to occur via activation of AKT signaling pathway. These findings support that hederagenin may be a potential drug for the prevention and treatment of depression.
