Glycyrrhetinic acid induces a protective autophagy in HepG2 hepatocellular carcinoma cells by activating ERK pathway

AbstractBackground: Glycyrrhetinic acid (GA), isolated from the Chinese medicinal herb named Glycyrrhiza, is a triterpenoid saponin with well-known anti-cancer effects. In this study, we aimed to study the effects and mechanisms of GA-induced autophagy in HepG2 cells. Methods: The anti-proliferative effect of GA in HepG2 cells was examined by MTT assay. The GA-induced autophagy was determined by monodansylcadaverine (MDC) staining, transmission electron microscopy (TEM) observation, and western blot analysis of lipid modification of light chain-3 (LC3) expression. The autophagy-related signaling molecules and extracellular regulated protein kinases (ERK) were examined by western blot analysis. The role of autophagy in GA-induced cell proliferative inhibition and apoptosis of HepG2 cells was assessed by MTT and western blot. Results: GA concentration-dependently inhibited the proliferation of HepG2 cells. It obviously induced autophagosomes formation, up-regulates the LC3II expression, and activated ERK pathway in a concentration-dependent manner. Whereas, pretreatment of mitogen-activated protein kinase kinase 1/2 (MEK1/2) inhibitors U0126 or PD98059, the GA-induced autophagy was remarkably reversed as supported by the reduction of autophagic protein marker LC3II expression. Additionally, the anti-proliferative and apoptotic inducing potential of GA were enhanced by pretreatment of autophagic inhibitors chloroquine or bafilomycin A1. Conclusion: To our knowledge, this is the first time to demonstrate that GA induces protective autophagy by activating the ERK pathway in HepG2 cells which shed a new light for the future application of GA as a new anti-cancer compound.
