Licochalcone A triggers autophagy by activation of endoplasmic reticulum stress in non-small cell lung cancer A549 cells

AbstractAim: Licochalcone A (LCA), a flavonoid isolated from the famous Chinese medicinal herb licorice, presents obvious anti-proliferation effects in multiple types of cancer cells. Herein, we’d like to determine whether LCA triggers autophagy in non-small cell lung cancer cells and the mechanisms involved. Methods: LCA-triggered autophagy was measured by the expression of the lipid modification of light chain-3 (LC3). SiRNA and pharmacological regulators were used to study the mechanisms of LCA-triggered autophagy. Results: LCA obviously induced the protein expression of LC3II, an autophagic marker, in lung cancer A549, 95D, and H1299 cells. LCA-triggered autophagy has been further confirmed by combined treatment with autophagic inhibitor choloroquine in A549 cells. Knockdown of autophagic related protein 7 by siRNA reversed LCA-triggered autophagy as evidenced by the decreased expression of LC3II. Besides, LCA induced obvious endoplasmic reticulum (ER) stress in A549 cells and the autophagic effect triggered by LCA was reversed by knockdown of C/EBP-homologous protein, suggesting that LCA triggered autophagy partially by activation of ER stress. Conclusion: As far as we know, our study demonstrated, for the first time, that LCA triggers autophagy in A549 cells via activation of ER stress.
