Abstract | Baicalein (BA), one of the major compounds isolated from the root of Scutellaria baicalensis Gerogi, is a bioactive flavonoid with various pharmacological effects. In this study, we evaluated the autophagy triggered by BA on human ovarian carcinoma HEY cells. We found that BA inhibited the proliferation on HEY cells after 24 h treatment as evidenced by MTT assay. Meanwhile, BA triggered the acidic vesicular organelle manifested by monodansylcadaverine staining and enhanced the expression of LC3-II in HEY cells. Inhibition of autophagy by chloroquine (CQ), an inhibitor of autophagy, the cell viability was obviously decreased whereas the expression of cleaved PARP was significantly increased, which means that BA triggered a protective autophagy. We also found an increase in phosphorylation of p38, ERK and AKT after BA treatment. Furthermore, pretreatment with U0126, an MEK inhibitor, obviously inhibited BA-induced activation of ERK as well as that of LC3-II. These results suggest that BA triggers a protective autophagy via activation of ERK, which might attenuate its anticancer effects in ovarian carcinoma cells. |
