Celastrol induces apoptosis, triggers autophagy and inhibits metastasis on ovarian cancer cells

AbstractCelastrol, a pentacyclic triterpene extracted from traditional Chinese medicial plant tripterygium wilfordii, presents obviously anti-cancer effects. However, the anticancer potential of celastrol on ovarian cancer, especially the paclitaxel-resistance ovarian cancer cells are still unclear. Herein, the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to evaluate the anti-proliferative effects of celastrol in paclitaxel resistant A2780/taxel cells and parental A2780 cells. After 24 h treatment, the IC50 of celastrol on A2780/taxel and A2780 were 3.31 ± 1.38 μM and 3.19 ± 1.51μM, respectively. Annexin V-PI staining assay indicated that celastrol remarkably induced apoptosis in both A2780/taxel and A2780 cell lines, and the expression of cleaved-caspase-3 and cleaved-PARP were increased while Bcl-2 was decreased. Besides, the autophagic protein marker phosphatidylethanolamine-modified microtubule-associated protein light-chain 3 (LC3-II) was increased after treatment of celastrol. In addition, celastrol inhibited cell migration and invasion on the HEY cells after 12 or 24 h treatment. Taken together, celastrol remarkably induces apoptosis, triggers autophagy and inhibits metastasis on ovarian cancer cells.
