Functional Genomics analysis of non-essential protein kinases.

AbstractProtein kinases accounts for 1.5-2.5% of the eukaryote genomes and are major regulators of cellular functions through reversible phosphorylation of protein targets. Thirty percent of human kinases are implicated in various diseases; hence, kinases are potential targets for many therapies. Using a Functional Genomics and Bioinformatics approach, we found that more than thirty percent of kinases are differentially expressed upon fungal infection of macrophages. These data implied that kinases played an important role in fungal pathogenesis. Using a collection of deletion mutants for 99 non-essential kinases in the model fungus A. nidulans, we undertake a high-throughput Functional Genomics approach to gain a global understanding of the physiological roles of individual kinases at the genome-wide level. Preliminary results of several kinase mutants already advocated probable pathways these kinases are involved. As kinase functions are highly conserved, our result will help to understand the physiological functions of kinases not only in an important model fungus, but also in other fungal species including fungal pathogens and perhaps even in some higher eukaryotes.
