Abstract | Aspergillus fumigatus is an opportunistic fungal pathogen that secretes an array of immune–modulatory determinants, including secondary metabolites (SMs), which contribute to enhancing fungal fitness and growth within the mammalian host. Gliotoxin (GT) is a SM that interferes with the correct function and recruitment of neutrophils, an immune cell that produces reactive oxygen species in order to fight A. fumigatus infections. We identified a transcription factor (TF) that is crucial for A. fumigatus oxidative stress resistance, virulence in an immunocompetent murine model of pulmonary aspergillosis, and for modulation of the murine immune responses. This TF regulates biosynthesis and self-protection the expression of gliT and gliF of the GT biosynthetic gene cluster through directly binding to the respective promoter regions in GT-producing conditions. Furthermore, this TF was shown to be crucial for GT biosynthesis and self-protection in A. fumigatus. Homologues of this TF were identified in other fungal classes, with the A. nidulans homologue also regulating the expression of a GT reductase-encoding gene, suggesting conservation of function for this TF in both GT-producing and non-producing species of Aspergillus. In conclusion, this work describes the function of an uncharacterised TF, important for GT biosynthesis and virulence, features that are also important in other fungal species. |
