Abstract | Crocin is an active ingredient found in saffron. Accumulating evidence suggests crocin is beneficial for neurodegeneration and Alzheimer’s disease (AD). Recently, neurodegeneration and AD are strongly correlated to neuroinflammation. Reactive astrocytes can release inflammatory mediators that contribute to neuroinflammation. This study aims to evaluate the protective effects of crocin in LPS-stimulated astrocytes. Mouse astrocytic cell C8-D1A was used as cellular model. Lipopolysaccharides (LPS) was used as an inducer to activate the astrocytes. Cells were pretreated with crocin followed by LPS stimulation. Cell viability, intracellular calcium influx [Ca2+]i, reactive oxygen species (ROS), nitric oxide (NO) and NF-ĸB signaling were evaluated. Our results showed that crocin was able to inhibit the LPS-induced cytotoxicity, ROS and NO production, and Ca2+ influx in C8-D1A. Furthermore, we showed that crocin suppressed NF-κB signaling by inhibiting the phosphorylation of IκB kinase (IKKα/β) and downstream phosphorylation and degradation of nuclear factor-κB (IκB). In conclusion, our data suggested that crocin could dampen LPS-induced inflammatory responses in astrocytes, and might be a potential candidate for treating CNS diseases. |
