AbstractPulsatilla chinensis (Bunge) Regel (P. chinensis) grows in northeast China such as Jilin, Liaoning, Shanxi, south of Gansu, etc. The chemical constituents of P. chinensis mainly include coumarin, lignin, flavonoids and triterpenoids, among which triterpenoids are the most abundant. Studies in recent years showed that some kinds of triterpenoid saponins exhibited great bioactivities. Cimicifuga dahurica (Turcz.) Maxim. (C. dahurica), a perennial herb grows in north of China. It was recorded in Chinese pharmacopoeia to treat with fever and pain. The 9,19-cycloartane triterpene glycosides in C. dahurica had considerable anticancer activity. Based on the previous studies on Pulsatilla saponin D, E4 and B4, further investigation of active chemical constituents in P. chinensis was explored. By using several chromatographic techniques such as microporous resin, silica gel, Sephadex LH-20, ODS, MCI and HPLC, 9 compounds were isolated from the water extract of the roots of P. chinensis. Based on UV, IR, NMR and HRESIMS, the structures of the 9 compounds were elucidated and identified as (8R,8’R)-Matairesinol 4,4’-di-O-β-D-glucopyranoside (YK-CD-1), (7R,8S)-dihydrodehydrodiconiferyl alcohol-di-9,9’-O-β-D-glucopyranoside (YK-CD-2), Eucomegastigsides C (YK-CD-3), 7,9′:7′,9-diepoxylignans pinoresinol-4,4′-di-β-O-D-glucoside (YK-CD-4), 3-O-β-D-glucopyranosyl (1-4)- β-L-arabinopyranosyl hederagenin 28-O-α-L-rhamnopyranosyl (1-4)-β-D-glucopyranosyl (1-6)-β-D-glucopyranoside (Y-L-I-5), cauloside G (Y-L-I-6), cauloside D (Y-L-I-7), clematangoside A (Y-L-I-8) and pulsatilloside E (Y-L-I-9). Among them, compound YK-CD-2 was firstly isolated from P. chinensis. The bioactivities of 9 compounds isolated from the roots of P. chinensis together with other 7 triterpenoid saponins from Pulsatilla koreana and 5 cycloartane triterpene glycosides from C. dahurica were detected. Compounds Cd-8, Cd-9, Cd-11 and Z-Ca-12 showed considerable cytotoxicity against MDA-MB-231 and MCF-7 cell lines. Compound Cd-8, Cd-9 and Cd-11 also increased ROS level and enhanced apoptosis in MCF-7 cells. Compound Cd-8 and Z-P-2 decreased NO production and IL-6 level on LPS-induced RAW 264.7 cells, which indicated their anti-inflammation potential.
