Rapid Regulation Of Oxidative Phosphorylation By Insulin In Human Embryonic Stem Cells

AbstractInsulin is an essential hormone for energetic metabolism in human. Besides its immediate promotion of glucose transportation, most insulin-dependent metabolic functions are carried out through transcription and translation processes that often take hours or even days. The direct role of insulin in oxidative phosphorylation (OXPHOS) is rarely reported, and its actions on a rapid time scale remain unexplored. At the same time, insulin is also essential for human embryonic stem cell (hESC) maintenance, but its role in stem cell metabolism is largely unknown. Here we show that insulin rapidly stimulates substrate-specific OXPHOS in hESCs. Pharmacologic and genetic manipulations reveal that insulin regulates OXPHOS through PI3K/AKT/GSK3 cascade within minutes before significant changes in the expression of metabolic enzymes occur. Intriguingly, we observed that insulin-dependent OXPHOS is hESC-specific, and it diminishes in differentiated cell types. Moreover, we noticed that insulin or GSK3 inhibition significantly suppresses the activation of caspases. Collectively, these results suggest that insulin regulates OXPHOS in a rapid mode, and insulin is continuously required to maintain full mitochondrial capacity in hESCs. Our findings provide molecular insights into the association between insulin signal transduction and mitochondrial energy metabolism, and highlight the necessity of insulin in the consistency and accuracy of metabolic analyses in intact hESCs.
