Abstract | Pyruvate is the central metabolite in the metabolic transition from glycolysis to TCA cycle during differentiation in human pluripotent stem cells. Many studies explored the functions of upstream and downstream metabolites of pyruvate, but the direct function of pyruvate is rarely discussed. In this study, we demonstrate that elevated pyruvate in medium potentiates and accelerates mesodermal differentiation induced by BMP4. Hyper-pyruvate shifts metabolic balance to oxidative phosphorylation and increases phosphorylation of AMPK and SMAD1/5/8. Activated AMPK leads to mTOR inhibition that then accelerates mesodermal differentiation. This study demonstrates that pyruvate not only is a key metabolite, but also serves as an important regulator of signal transduction pathways underlying cell fate determination in stem cells. |
