Abstract	Tubes are a fundamental unit of organs in all multicellular animals, such as lung, digestive tract, kidney and blood vessels. Polarization of lumen epithelium serve as a foundation for tubular morphogenesis. Failure to establish or loss of maintenance of lumen polarity disrupted biological development. During our previous study, we found that one of the RNA-processing gene can cause apical polarity defect through RNA-mediated interference (RNAi) based screen. Intracellular mRNA localization is a kind of post-transcriptional control of gene expression and involved in mRNA turnover and translation to acquire the local protein production. We hypothesis that RNA-processing may have influence on epithelial polarity through RNA quality control which affect the localization of their encoded protein of polarity complex or affect polarity complex to receive signal from membrane localization protein. To figure this out, we need to verify in this way: 1. A targeted RNAi mediated tubulogenesis screen to RNA processing related genes to acquire the potential pathway. 2. Elucidate the mechanism of how RNA-processing affect epithelial polarity and tube morphogenesis. Many important human organs are simple tubular structure during embryonic stage and develop into complex structures as they grow up with different functions. The study of tubulogenesis mechanism help to understand the pathology of some developmental diseases which caused by abnormal tube formation and hence give indicates to the treatment.
