Abstract	Localized mRNAs in subcellular compartments provides spatial and temporal protein expression which is an effective way to create structural and functional asymmetries in cells. Over the last few decades, a great amount of evidence has shown that cis-elements on the RNA sequence, RNA-binding proteins and the transport motors are critical to RNA translocation. Recent researches have furthered our understanding of how the mRNA-protein components localize to certain cytoplasmic domains, by new and improved techniques which describe the connection between mRNAs and trans-acting factors, dynamics of mRNA trafficking and physiologic effects of localized mRNA in cells. The innovative imaging methods have provided vast knowledge on mRNA localization, as an example, the trafficking and localization patterns of mRNAs in specific cytoplasmic domains have been illuminated by observing single molecules in unicellular organisms or in tissues. Here we focus on cellular mechanisms of delivering the RNA to its destination which include recognition of localization elements; function of regulatory RNA-binding proteins; and role of molecular motors and cytoskeletal elements on mRNA localization. Then we discuss how visualize mRNA technologies have furthered our vision of endogenous mRNA localization in various types of cells and tissues. These involve microinjection of labeled transcripts into living cells, in situ hybridization by using labeled antisense probes and complemental transgenic methods for labelling endogenous mRNAs using fluorescence protein components. These improvements of imaging techniques provide the potential to renovate our knowledge of the subcellular mechanisms of mRNA localization.
