Abstract | The nod-like receptor 3 (NLRP3) inflammasome is a caspase-1-containing multi-protein complex that controls the release of interleukin-1β (IL-1β) and plays important roles in the development of inflammatory diseases. Our previous study reported that HSQ14, a cycloartane triterpenoid isolated from the whole plant of Actaea vaginata (Ranunculaceae), possesses anti-inflammatory effect on lipopolysaccharide (LPS)-treated RAW264.7 macrophages. However, the underlying mechanism hasn’t been clarified clearly. Herein, we investigated the effect of HSQ14 on mitochondrial integrity and NLRP3 inflammasome activation in vitro and in vivo. HSQ14 alleviated LPS-induced inflammation in RAW264.7 cells through activating SIRT3-mediated autophagy. Furthermore, HSQ14 inhibited the activation of NLRP3 inflammasome by enhancing mitophagy and ameliorating mitochondrial damage. In addition, upregulation of AMPK/ULK1 signaling pathway accounted for HSQ14-driven autophagy. HSQ14 treatment alleviated LPS-induced acute inflammation in C57BL/6 mice, indicating by the inactivation of NLRP3 inflammasome and suppression of pro-inflammatory cytokines, including IL-1β, tumor necrosis factor-α, IL-6 and monocyte chemoattractant protein-1. In conclusion, our results demonstrated that HSQ14 might be potentially applied for the treatment of NLRP3-associated inflammatory diseases. |
