Abstract | Penthorum chinense Pursh (P. chinense), a traditional Chinese medicine, is used for liver protection and treatment of liver diseases, including hepatitis B, hepatitis C and alcoholic liver damage [1]. Recent research reported the hepatoprotective effect of extract of P. chinense on alcohol or t-BHP induced liver injury [1-3]. In the current project, we employed the model of t-BHP induced oxidative damage on normal human HL-7702 cells, and attempt to identify fractions with controllable constituents, and/or compounds from P. chinense, which possess potent hepatoprotective property. Several fractions were found to significantly protect hepatocytes from oxidative injury. Furthermore, 10 compounds were identified from the most potent fraction. Among them, quercetin, a well-known common flavanone widely distributed in different plant, showed protective effect on the cell model. Further investigation demonstrated that intracellular level of reactive oxygen species (ROS) was significantly decreased by treatment with quercetin in a dose dependent manner. Additionally, quercetin was effective in protecting hepatocytes from being injured by t-BHP through modulation of the expression of apoptosis related proteins, such as cleaved-PARP, cleaved caspase-7, cleaved caspase-9, Bcl-2 and Bcl-xL. In summary, it was deduced that the bioactive compound, quercetin, exerts its novel protective effect on t-BHP induced hepatocytes injury through decreasing the intracellular level of ROS and modulating the expression of apoptosis related proteins. The current work sheds light on the chemical principles of hepatoprotective effect P. chinense, and supplies basic data to further develop this traditional herbal medicine as drug or healthy food. |
