Abstract | Obesity is one of the most common metabolic diseases worldwide, which is characterized by the excessive accumulation of triglyceride in adipocyte. Recent evidences demonstrated that the lipid droplets (LDs) in adipocytes are selectively degraded by lipophagy to release free fatty acids and maintain energy balance. Meanwhile chaperone-mediated autophagy (CMA) regulates the mobilization of intracellular lipid by degrading the LD coating proteins. Sirtuin 3 (SIRT3), a mitochondrial NAD+-dependent deacetylase, plays a crucial role in regulating energy metabolism. The role of SIRT3-mediated autophagy in adipocytes is controversial. In this study, a SIRT3 overexpression (SIRT3OE) cell line was generated to investigate the role of SIRT3 in autophagy and lipid accumulation in 3T3-L1 adipocytes. Our results showed autophagy is dynamically regulated during adipocyte differentiation which coincides with SIRT3 expression. In fully differentiated adipocytes, SIRT3 induced autophagy, mainly on LDs, through activating AMP-activated protein kinase (AMPK). SIRT3 overexpression decreased the lipid accumulation and increased the breakdown of LDs through lipophagy. SIRT3 overexpression elevated perilipin1 degradation on lipid droplets, which in turn increased lipid mobilization. When administrated with Compound C (an AMPK inhibitor), the effects of SIRT3 on lipid accumulation and CMA were almost abolished. Our findings indicate that SIRT3 attenuates lipid accumulation in mature adipocyte by enhancing CMA and lipophagy through activating AMPK, and suggest that SIRT3 activation can be a potential therapeutic choice for the treatment of obesity as well as its related metabolic disorders. |
