Α-Mangostin ameliorates aging related adipose tissue inflammation and metabolic disorders | |

Abstract | Obesity in the elderly individuals is increasing at an alarming rate; and low-grade inflammation in adipose tissue contributes to the onset and development of metabolic disorders, such as insulin resistance and type 2 diabetes. α-mangostin was the main xanthone in mangosteen (Garcinia mangostana), which has been used in traditional medicine for treatment of various inflammatory diseases. The present study was designed to test the hypothesis that α-mangostin could alleviate age-related adipose tissue inflammation. Firstly, α-mangostin ameliorated lipopolysaccharide (LPS)-induced inflammatory responses in RAW264.7 macrophages, and tumor necrosis factor α (TNF-α) mediated inflammation in 3T3-L1 adipocytes, through inhibiting the mitogen-activated protein kinases (MAPKs) and nuclear factor-κB (NF-κB) activation, and promoting sirtuin3 (SIRT3) expression. In addition, α-mangostin blocked RAW264.7 macrophages migration towards 3T3-L1 adipocytes in cell co-culture system. Next, α-mangostin attenuated inflammatory responses in LPS-induced acute inflammatory mice model. Lastly, we recruited a cohort of young (4 months old) and aged (20 months old) mice, and found α-mangostin reduced aging-related inflammation in adipose tissue. α-Mangostin reduced the levels of pro-inflammatory cytokines and chemoattractants, inhibited macrophage infiltration into adipose tissue, and reversed the age-related shift towards pro-inflammatory macrophages, through suppressing the activation of MAPKs and NF-κB pathways. In aged mice, α-mangostin decreased the mass of epididymal fat, improved insulin sensitivity and ameliorated liver steatosis. Overall, the above results demonstrated that α-mangostin might represent a novel therapeutic agent for treatment of aging-related chronic adipose tissue inflammation and metabolic disorders. |
