Functional study of the Cak1 kinase in Aspergillus nidulans

AbstractCyclin-dependent kinase (CDK)-activating kinase (CAK) is a highly conserved protein kinase that activates cyclin-CDKs. While the main function of CAKs is in the control of cell cycle, CAKs have been shown to also regulate transcription in a number of organisms including Saccharomyces cerevisiae. For example, Cak1 in Saccharomyces cerevisiae phosphorylates CDKs such as Bur1, Bur2 and Kin28, which play important roles in the RNA polymerase II transcription process. While S. cerevisiae Cak1 has been extensively studied, the role of Cak1 in Aspergillus nidulans is largely unknown. In this study, we are interested in finding out whether A. nidulans Cak1 also play a role in the transcription process. While the cak1 gene is not essential in A. nidulans (De Sousa et al., 2013 Plos One 8(3):e58008), the cak1∆ mutant has a strong growth defect. Moreover, when growing on solid media, the cak1∆ mutant produces many sectors (i.e. suppressor mutants), which might simply be a consequence of severe growth defect. The suppressor mutants display diverse phenotypes such as variable growth rates, different levels of conidiation and sexual development, etc.. Therefore, the overall cak1∆ phenotypes have made the study of this kinase extremely difficult. To better study the role of Cak1, we have created analog-sensitive (as) Cak1 kinase strains, which allow conditional inactivation of the Cak1 kinase activity by using allele-specific kinase inhibitor analogues such as 1NA-PP1. Genome-wide transcriptional profiling was performed on the as-Cak1 as well as the cak1∆ strains in order to uncover the physiological and molecular roles of Cak1 in A. nidulans.
